1> Firstly prepare for the five-time M9 salty solution.
M9 solution recipe (1L):
64g Na2HPO4, 15g KH2PO4, 2.5g NaCl, 5.0g NH4Cl. Add the ddH2O to dissolve all the solute till 1L. Then have the M9 salty culture sterilization.
2>Secondly prepare for the M9 culture
200ml M9 salty solution, 2ml 1mol/l MgSO4, 100µl 1mol/l CaCl2. Add ddH2O till 1L. Then get the M9 culture sterilized.
